I have done 50/50 glue and water, then sanded smooth... messy.
I have heard of drywall compound?
Anything else?

I've never done it (edge sealed MDF), but I've seen a lot of tips and tricks posted online
- Bondo
- Thinned glue
- Drywall compound (use the 'hot mud' stuff - dries harder and faster)
- latex caulk
- various versions of plumbers putty
- BIN primer (from Zinsser)

I like Bondo. It's fast, durable when dry, and terrific to sand.

i done 2 small mdf projects and sealed them with this:

2 coats of the primer around routed and cut edges was all it took,
here's a link to what the finish looked like after the primer/sealer was put on and the finishing coats:

That looks like it's worth a try! I haven't seen it before.

Any oil based primer is fine.

Enduro white undercoat. It's a block-out primer. Sands very easy to a fine powder.

We just shoot the edges with oil-based primer like dwaker said... sand and repeat...normally 2 coats depending how it looks.
